Letters are the Latin-script letters above the Alphablocks. They are small black symbols encased in the characters’ domes. Their function appears to be to identify which Alphablock they are, and what sound they make/what their name is. There are 2 types of symbols: CAPITAL/UPPERCASE/BIG letters, and lowercase/small letters/minuscule letters.

- The letters are almost always in lowercase, although their capital letter-variants have made appearances over the years, such as:
- A in the Alphablocks logo.
- E (third) and T (third) in "Fred".
- H (third) in "Book".
- P (third) in "Sleep" and "Outlaw".
- I (third) in "Mine"; although it seems to be the same as the capital I from the Heinemann Roman Bold font.
- N (third) in "Making Friends".
- B (fourth), Y, and T (third) in "The Case of the Missing Blocks".
- S (third) and E (second) in "Letters to Santa".
- U, F, and O in "UFO".
- All of the capitals in the top rows in "Band Together"; although they aren't in the Alphablocks themselves, there are seen in signs and in the map alongside the regular lowercase letters.

- Arial Rounded - the text in the storybook in "Book"; this font has redesigned letters to match the Alphablocks' original letters, like "a".
- Blocks Universe font - original font used by Alphablocks, Ltd. for modern merch, like annual.
- The appearance of the uppercase letters are almost never consistent. A notable example is the capital J.
- In "Book", it is shown that Alphablocks can swap out their letters with other ones and transform into the Alphablock of that letter.
